Load the CellexusBag or HybridBag The cell culture bag is loaded into either of the two CellMaker Enclosures. Access flaps in the enclosure make it simple to locate the tubes and filters.

Close the lid The CellMaker Enclosure is closed and secured with safety interlocks, which means that it cannot be opened during operation. Obvious perhaps but an important safety and security feature.

Connect the cell culture bag The bag is connected to the CellMaker Controller and the media pump, via a sterile Pall® filters1 which can be provided pre-sterilised as part of the bag, or can be added immediately prior to use - these filters may be used several times and withstand several autoclave cycles to keep running costs to a minimum.

|
Programme the CellMaker Controller The CellMaker Controller can be programmed to deliver any flow rate from 1-10 litres per minute. Flow can be in pulses with a duration from 1 second to 10 minutes, with intervals from 1 second to 10 minutes in duration.

|
Set the pressure The pressure inside the bag can be set at low (atmospheric), medium or high. This equates to a pressure of up to 50mBar or 0.75 psi. This can dramatically increase gas transfer from the top space into solution and reduce foaming - by analogy think of an unopened can of soda.

|
Load media through a Pall® KleenpakTM media filter1 Sterilise the media using in-line filtration as it is added to the bag or fill with pre-sterilised solutions from separate containers or bags using the Pall® KleenpakTM connectors. |
|
Inoculate vessel through the culture/sample ports Culture/sample ports are included to facilitate easy inoculation and removal of samples from the culture.
|
|
Adjust the flow rate The flow rate is adjusted to provide sufficient mixing and aeration. An oxygen sensor using our non-invasive WIDGETS can be used to measure the dO2 of the culture and pH (HybridBag).

|
Harvest culture through the media addition port The cell culture bag will empty under gravity, internal gas pressure or via a peristaltic pump as required.
|
|
The CellMaker systems have a unique bioreactor geometry.
